Clausenalansine A – first reported in 2013 – is a recent addition to the family of carbazole alkaloids, which have long been at the center of considerable scientific interest, owing to their important and diverse range of biological activities. This compound was isolated from the wampi fruits (Clausena lansium) and displayed remarkable neuroprotective activity. Recently, the group of Professor Hans-Joachim Knölker from the Technische Universität Dresden (Germany) reported the first total synthesis of the title compound, which could potentially enable medicinal chemistry campaigns aimed at the design, preparation and screening of synthetic analogues of this alkaloid. The study confirmed the structural assignment of the natural compound and allowed a detailed X-ray study of the crystal structure of clausenalansine A.

Professor Knölker said: “We have developed an efficient synthetic access to the neuroprotective pyrano[3,2-a]carbazole alkaloid clausenalansine A and thus, our work may pave the way to potential novel lead structures for the treatment of neurodegenerative diseases.”
